DSC (Differential Scanning Calorimetry)Transition TemperaturesDSC provides rapid and precise determination of a variety of transition temperatures needing only a minimum amount of sample. Transition temperatures can be determined as well as resulting enthalpies (exothermic or endothermic). Measurable transitions include melting, glass transitions, crystallization, solid-solid transitions and mesophase transitions. Polymorph AnalysisPharmaceutical materials often exist in multiple crystal forms. These have the same chemical structure but a different crystalline structure which can result in significant differences in physical properties such as solubility, bioavailability and storage stability. DSC is the prevalent technique for the detection of polymorphism. Using heat-cool-heat techniques, the stability of various polymorphs can be identified and quantified. Amorphous Solids and Crystallinity QuantificationPartially or completely amorphous solids exhibit a glass transition temperature and can exhibit a melt transition. Knowledge of glass transition temperatures is important for pharmaceutical stability storage and can be precisely determined with DSC. Changes in enthalpy of fusion detected by DSC, in comparison to a fully crystalline sample, can be used to determine the degree of crystallinity in an amorphous sample or vice versa. Decomposition and Residues Analysis
Decomposition of organic materials and knowledge of eventual residue content is an important tool in product design and performance. Changes in decomposition temperature due to fillers or additives could be critical for the evaluation of new product candidates. High-resolution TGA allows the precise determination of decomposition temperatures and accurate quantification of residues after decomposition. Volatiles Quantification Determination of absorbed, bound or occluded moisture and organic volatiles is an important analysis for product performance. Further, pharmaceutical scientists routinely characterize the thermodynamic parameters of drug candidates including solvates and hydrates. TGA measurements allow rapid and precise determination of weight losses due to moisture or loss of crystal water. Reaction Kinetics and Activation Energy Knowledge of the kinetics of phase changes, conversions or decompositions is an important tool in assessing physical stability with temperature. Elucidation of conversion mechanisms is possible using the activation energy of the event. Modulated TGA provides not only the weight loss during the transition but also the associated activation energies in a single experiment. |
